Material Identification： 

Character identification: This product is composed of a worm body and a sub-seat that grows from its head.

The worms resemble silkworms with a length of 3 to 5 cm and a diameter of 0.3 to 0.5 cm.

The surface is earthy yellow, dark yellow or yellow-brown, with obvious ring patterns, 20 to 30, reddish brown on the head, and fine ring patterns near the head.

There are 8 pairs in full, 3 pairs near the head, 4 pairs in the middle and 1 pair near the tail.

The texture is brittle, easy to break, the section is slightly flat, pale yellowish white.

The pedestal is slender and cylindrical, with a length of 4 to 7 cm and a diameter of about 0.3 cm. The surface is dark brown to brown with fine longitudinal wrinkles, and the upper part is slightly enlarged.

The quality is flexible and the cross-section is almost white.

Slightly fishy smell and slightly bitter taste.

The ones that are complete, yellowish, plump, and stubby are better.

Physical and chemical identification 1. Take 1g of powder, dissolve impurities with ether, extract with chloroform, filter, evaporate chloroform from the filtrate, add 2 drops of glacial acetic acid, then add 2 drops of acetic anhydride, and finally add 1 to 2 drops of concentrated sulfuric acid.

Brown-yellow→red-purple→dirty green.

2. The chloroform-extracted powder is extracted with 20% ethanol under reflux and concentrated to an appropriate amount.

(1) Take 1ml each of the concentrated solution, divide them into 2 test tubes, add a few drops of dilute hydrochloric acid to one tube, and then add a few drops of the bismuth potassium iodide test solution. After standing for 10 minutes, a yellow flocculent precipitate is produced.

The iodine-potassium iodide test solution was dripped into another test tube, resulting in turbidity.

(2) Thin layer chromatography: Take the sample prepared above, the adsorbent is silica gel G, and the plate is made with 0.3mol/L boric acid.

The developing agent is n-butanol-acetic acid-water (2:1:1).

The span is 17cm.

Color developer: a is 0.1% potassium periodate, b is 0.5% benzidine alcohol solution.

Disperse liquid a for 5 times, spray it on the plate, dry it, and spray liquid b to show white spots on a blue background.

The UV spectrum was measured on Shimadzu UV-210A UV spectrophotometer.
